A Joseph Hodgkins married Ann Adams 11th Feb. 1787 at St Peters Aston
assuming he was literate would he have signed the register as early as this date?

I have a signature of a Joseph Hodgkins ( w ) marrying a Hannah Mattocks ( w )
5th June 1809 in St Peters Wolverhampton and I and wondering if it is the same
person, thought maybe the sig. could provide a clue.

any help would be appreciated,

Yes, if the person was able to sign their name, it should be in the original marriage register in 1787.  The entry would look something like this one, from 1755:
