I know very little about DNA, so some advice needed please.
 Joe Bloggs (father unknown) married twice he has a daughter and son with each wife.Joe is now deceased.
Would it be possible for one of those children to do a DNA test and attempt to find a connection to their unknown grandfather. Also would it be best for a male or female to do the test or would it not make a difference. thank you for reading.

Yes, it would be possible for either to do it, but the male would probably be better because he could also do the Y-DNA test in addition to the autosomal test.  This kind of explains the two tests - https://www.familytreemagazine.com/premium/autosomal-vs-ydna-testing/

The best way to test is with Ancestry as they have over 5,000,000 testers - so you are more likely to find matches.
I would test both children this would help confirm your matches when you get them.
The y-dna male test is with FTDNA  and would be a last resort if you didn't get your results through Ancestry. It is a different kind of test and is probably not so appropriate in this case.
