Googling William's name reveals:

His son, George Coombs Poole, grocer, became bankrupt in 1839 and William was also named.

Jo Draper's history of Lyme Regis says that William was a wine merchant and built a large house, Summer Hill, in 1819 which he let out to visitors (now demolished but photo online).
 
William is on the list of Dorset voters in 1807, so must have been wealthy by then:

There appear to have been Poole landowners in Lyme Regis for a number of generations. The Dorset History Centre has a range of documents on William, so it may be worth contacting them.

You will also see various other Pooles owning land who are probably related.

There is also a William Poole, clothier of Lyme Regis, who left a PCC will in 1717. There is no evidence yet that he is related but it suggests that the family had been trading in the town for some time.

On 7 Aug 1775 the Sherborne and Yeovil Mercury states that William Poole was importing wines:

Given that your William would only have been about 13 at the time, this could suggest that William's father was also William and also sold wines.

This history of Lyme Regis, written in 1823, has a Mr Poole as one of the subscribers and says that Cumberland House is owned by Mr Poole but is 'now occupied by Colonel Williams':

There is a road in Lyme Regis called Poole's Court which I had always assumed was something to do with the town close to where I live; but perhaps it was named after your ancestor as the towns are at opposite ends of the county.
