While checking emigration record of cushen sisters who left queensland april 1912 i noticed that opposit the line of Margaret Cushen there appears to be a clear stamp of 1940. What is the significance of this when the actual record is from 28 years previously (ie) 1912

It looks like that may be in relation to a subsequent application for naturalization.  This explains about how they checked previous passenger lists to see if they could find the person applying for naturalization and how they marked it - https://www.jewishgen.org/infofiles/manifests/occ/

Yes, I think they were checking to see if the person came when they said they did.
