There was a Stannus Square Lisburn in 1870's and a linen mill there.

Hmm could well be Stannus. I see there's a Stannus Place in Largymore, Lisburn.

Problem is they're in Belfast, not Lisburn.

I would still go for ‘H’ as the initial letter.

Perhaps the square was named after Dean Stannas who was a bigwig there .....
Cannot find an old map of it.

If you look at the whole page the 'St' in Stevenson (next entry after William Jefferson's birth) is different, whereas the 'H' in Holmes (entry before William's birth) looks the same so I think is says Hanna's SQUARE.
Marriage of William's mother Mary Jane Jefferson (21 July) gives almost the same address as on birth registration (born 26 May, registered 14 June) - Hanna's? COURT-
