What method could I use to prove this?

Very difficult. I think if it were my ancestors, I'd explore the parents for any siblings and any reasons for going to Tranmere.  Were there any will, etc. ?

The alternative  (scientific method) is to try your best to disprove that he was yours - chase up this person and find that he didn't move to Tranmere  If you can't find him in Anglesey/elsewhere, then he might well be your ancestor.
