Putting it all together, James WELLS & Eliza GREEN along with their children Robert, James, John & Eliza came to Canada aboard the Sisters shortly before or on 4 Aug 1851 when both Elizas died in quarantine.

James may have remarried, the 1861 census of Glenelg Township, Grey County has James 55, Mary 53 and John 12.

John Wells married Margaret Charlton 6 Sep 1875 Bentwick Township, Grey County, Ontario.  Age is out by a few years, but I think this is the right John.

John Wells death 30 Oct 1930, note daughter was informant and mistakenly entered her parents names.

From the British Parliamentary Papers, 1852 XXXIII (1474)
Report of migration to Canada 1851.

"SISTERS" ( dated 25th July -assumed to be arrival ) from London 1851, 8 Passengers paid for by Lord Corrington (believed to be Carrington) , 94 Passengers paid for by poor law Unions at a total cost of £74 10s ( Wayland Union documents to be investigated on line)
destination Canada West and 37 passengers to Buffalo and Chicago.
As a back drop.
9677 people from England migrated to Canada in 1851, their numbers swollen by an additional 59 births en route, but 210 passengers died, a further 51 died in quarantine. ( Cholera and Smallpox being a particular problem in Europe )
The average journey time  England  - Canada  41 days.( this seasons average was 44.5 )

A James WELLS married a Mary LOCKHART in Wentworth Ontario in 1852, is this Roberts father? This would hopefully detail James parents names, and I could identify his baptism in England.

The marriage was from Vol. 18 Newcastle District 1850-1854
Unfortunately the bride and groom, date of marriage and witnesses - seemingly unrelated and whether it was licence or banns is all the information recorded.

Do you have the 1837 marriage registration for James and Eliza as that would give father's name.
