I don't know abut any sisters of John Cameron I'm afraid. My research so far as mostly been on direct ancestors. Just starting to branch out into siblings of GG GdPs now.

I wonder if by 'wife' they just meant the person was married?

As suspected the banns were called in both parishes and the actual marriage followed soon after in one. IGI does not give accurate details as you see below.

Cameron & Innes
John Cameron, Confectioner, Residing in the Town and Parish of Kirkcaldy
and Elizabeth Innes, Residing in No. 13 Carnegie Street in this Parish, Daughter
of John Innes, Surveyor of Customs in Greenock have been three several times
duly and regularly proclaimed in order to Marriage in the Parish Church of St
Cuthberts and no objections offered. Married on Twelfth day of February, 1839
by the Reverend Hugh Mcleod, Minister of the Gaelic Church in Edinburgh.

Following-up Elizabeth Innes' father, John Innes ..
it looks as if Elizabeth was a birth from a first marriage c. 1810 in Tain, Ross and Cromarty (or illegitimate), and then John went to Greenock where he married Agnes Service.
There is no record of an Elizabeth Innes birth/christening c. 1810 in Tain to John Innes in either GRO or IGI. She may have been christened with her mother's name, if illegitimate, or not recorded at all.

However a christening record for John Innes provides another Great x n Grandfather plus his parents as Great x n+1 Grandparents !
