1) What was the name of JAMES DARRINGTON's first wife and did they have children.

2) Above John Hart married either Mary Anne Darlow or Emma Jacques in D1871 Caxton. What became of them?

Looks like he was an unmarried servant in 1851, and we know he was a widower in 1861, so he married and his wife died between 1851-1861.

From his second marriage, we also know that his father was William Dorrington.

Possible first marriage:

Spouse: Hannah Holden
Father: William Darrington
Age:   29
Birth Date: 1825
Marriage Date: 1854
Marriage Place: Godmanchester, Huntingdon, England

A Hannah Darrington of Gamblingay died in 1860, aged 27 (born c.1833).

Elizabeth (1856), likely burial in Gamlingay in 1857.
William (1858), likely burial in Gamlingay in 1861.
Jacomina (1860), likely burial in Gamlingay in 1860.

I don't think so

If your question is related to Harry Darrington who predates the marriage I think he's Caroline's son. I can;t find a Henry or Harry Darrington birth that would fit that's local (added there's one in Bedford, but he can be clearly found with family in 1871)

I agree with Mabel; no reason to think James married again between Hannah and Caroline.

To answer the question about John Hart's marriage, the 1871 marriage in Gamblingay is to Mary Ann Darlow, daughter of William Darlow. It looks like they moved to Shoreditch in London:
