Can anyone please tell me if there was such a job as supervisor of excises and what it would have entailed.

Part of Customs & Excise, some staff records are retained at the NA:

Presumably he was an Excise officer who had been promoted - there appears to have been a specific rank of 'Supervisor' - probably for the local office, whichever one that would have been.  You'll probably find him in local trade directories.
http://www.mkheritage.co.uk/tdlhs/Organisations/names_1849.html - here's an example, 1849 directory for Towcester lists Mr John W. Johnson as the Supervisor, Excise Office.

Smugglers of bootleg alcohol were the main opponents of the excise officers:
