Since his birth is before the start of full civil registration (1864), you will first need to establish where in Ireland the family lived at the time, as well as the family religion so that you can research parish records (if they have survived)

Nowhere on the Ancestry records do we see anything other than Irealnd as his place of birth. However, there is a link above which may connect you with someone who haas further informaton.

Exact birth place in Ireland, 1843.? Father William, born 1809, Tewkesbury, Gloucester.

William was a Sergant, but I don,t know with which Force, Army or Navy. It was on the marriage certificate as father to Nicholas.(Gloucester records).  Nicholas married Sarah Ann Dart in 1877, he died 1930.

Not sure if this is your man but from Woodard Database in Tewkesbury there is this from Tewkesbury Register Newspaper September 1921:-
Birthday Celebrations - Messrs. Nicholas A Mann Of Tewkesbury And William E Mann Of Rock Ferry, Cheshire, Celebrated Their 79th Birthday.  Both Cabinet Makers. Joined 5th Lancashire Artillery In 1865 Both Ulster Loyalists.  Grandfather Former Nicholas Mann Of Tewkesbury.  Brother, John Collins Mann, Oifficial Gt. Northern And Ireland Railway

and in November 1888:-
Death - 04/11/1888 at Newry in Ireland: Robert Mann, late Sgt. Major of Essex Regiment aged 58.  Eldest son of late Sgt. Major William Mann also ex-56th Regiment.  He was a Crimean War Veteran who had been port of Sebastopol and the Indian Mutiny; he had 6-7 medals;  Nicholas Mann was still in Tewkesbury and had helped found the Volunteers with Captain Moore.  He was the grandson of Nicholas Mann

The above link tells us that the 56th regiment of Foot - The Essex Regiment - was posted to Ireland in 1842.  That would be consistent with the birth date in Ireland of 1843.

So, it seems that Rootshacte has once again come up trumps and we know why there was a birth in Ireland, the rank and service of William Mann. What you will not discover unless you can examine the regimental papers is exactly where Nicholas was born. He may have been baptised by the regiment's chaplin.
